Relevance Giant isoforms, encoded by Nesprin-1 (Syne1) and Nesprin-2 (Syne2), are multifunctional actin-binding and nuclear-envelope-associated proteins belonging to the spectrin superfamily. Nesprins play pivotal roles in maintenance of nuclear envelope integrity, nuclear positioning, and anchorage to the cytoskeleton and the centrosome. Although compelling evidence underlines nesprin's roles in several human diseases that range from cerebellar ataxia to dystrophy-like phenotypes; the underlying molecular mechanisms remain elusive.
