The family of D-type cyclins (D1, D2, and D3) regulates the G1/S-phase transition. D cyclins bind and activate cyclin-dependent kinases (Cdk4 and Cdk6) to phosphorylate the retinoblastoma (pRb) protein, and through titration of the Cdk inhibitors p21Cip1 and p27Kip1, the cyclin D–Cdk4/6 complexes activate cyclin E/Cdk2. Genetic analysis studies in mice have demonstrated an essential role for cyclin D1 in normal development of the retina, components of the nervous system, and terminal alveolar breast bud development.
