Rock2 (Cow):ER GFP Plasmid contains kinase domain along with part of the coiled–coil region of bovine Rock2 (residues 5–553) attached to the hormone‐binding domain of the mouse estrogen receptor (hbER) at the carboxyl‐terminus and enhanced green fluorescent protein (EGFP) at the amino terminus. Rock2 (Cow):ER GFP Plasmid facilitates the study of Rock signaling in a more regulated manner by offering a conditionally active form of Rock2 through its fusion to the hormone‐binding domain of the estrogen receptor (ER). Activation can be achieved using the estrogen antagonist 4‐hydroxytamoxifen (4‐HT). Ultimately, Rock2 activation leads to phosphorylation of proteins involved in the regulation of the actin‐myosin cytoskeleton, generation of pronounced actin stress fibers, and changes in cell morphology. Rock2 (Cow):ER GFP serves as a good complement for small‐molecule Rock2 inhibitors. |
