Relevance The human embryo kidney cell line HEK293T modified for use as a reporter cell line for Activin and TGF-beta signalling. The cell line stably expresses CAGA12-Luc, a Smad3/Smad4-dependent reporter containing 12 copies of the CAGAC sites (AGAC motif underlined) from the PAI-1 promoter. This is a Smad3/Smad4-dependent reporter, although it also binds complexes of Smad2 exon 3 with Smad4. The cell line also contains a Renilla reporter driven by the thymidine kinase (TK) promoter to act as an internal control.
Production Details Made by transfecting the human embryo kidney cell line HEK293T with CAGA12-Luc with pRetroSuper for puromycin resistance and then TK Renilla with pCMV-Bsd for blasticidin resistance.
