Flow cytometry histogram: CHO-CEACAM1 cells were stained with purified his tagged human rec. HopQ (5 µg/ml) followed by mAb 2B6 (thick line) or isotype control mAb (thin line), followed by goat anti mouse-FITC. After washing samples were analyzed by flow cytometer. Dead cells were discriminated by staining with propidium iodide (PI) and excluded from the analysis.
Indirect Enzyme-Linked Immunosorbent Assay (ELISA) Indicated his-tag antigens or control antigen were coated onto wells. After blocking with 1% BSA/PBS His-tag mAb 2B6 (3 µg/ml) was added. After washing HRP-coupled anti mouse Ig pAb (goat) was incubated, washed and visualized by TMB detection.
Two different His tagged proteins were separated in a 7% Tricine gel, blotted to a nitrocellulose membrane and after blocking with 1% milk/PBS incubated with 3 µg/ml mAb 2B6 diluted in blocking buffer over night at 4°C. Subsequently, membrane was washed twice, incubated with HRP coupled goat anti mouse antibody, washed and visualized by ECL detection for 1 sec.
