Treatment of the cells Discard the culture medium and replace by 100 ul medium containing -30ng /ml murine IL-17A -100 ng murine IL-17F If you want to test inhibition activity of mAbs, préincubate a mixture of IL-17 and serial dilutions of the mAbs ( from 1000ng/ml to ± 1 ng/ml) in separated wells (min 30 min, usually around 4H) before adding to the 3T3 cells. Incubate the cells + mAbs + IL17 min 24H, preferably up to 40H. Collect SN from each well and store frozen till IL-6 Elisa. IL-6 ELISA – dilute SN 1/10 to perform the murine IL-6 ELISA. PS. Some FCS batches induce rapidly a shift in the 3T3 cells that become either insensitive to IL-17 or stop IL-6 secretion in response. Prepare quite a large batch of frozen vials as soon as possible. |
