Relevance This mouse models allows for researchers to track an visualize endogenous exosomes in real time. A NanoLuc reporter is fused to the exosome surface marker CD63 and expression is under control of a cardiomyocyte-specific promoter and conditional for tamoxifen.

Mouse Genetic Background/Cross History In order to achieve a controllable exosome tagging, a dSTOP sequence flanked by two loxP sites was introduced upstream of CD63NanoLuc, and the expression cassette was driven by cardiomyocyte-specific α-MHC promoter (Fig). Linearized DNA containing αMHC-STOP-CD63NanoLuc cassette was used for pronuclear microinjection to generate the founder of the transgenic strain.
