Monoclonal antibody directed at large T antigen of SV40, valuable for investigation into other SV40 tumour antigens and polyomaviruses. Background and Research Application Anti-SV40T which binds to the large T antigen of the Simian oncogenic polyoma virus (SV40). Both the Simian virus (SV40) large T and small T antigens are encoded by the early region of the SV40 genome. The SV40 large T antigen is an early protein that interacts with the p53 and Rb tumour suppressor proteins. Large T is capable of inducing malignant transformation within cells, predominantly due to its disruption and inactivation of the 105-retinoblastoma and p53 tumour suppressor. This causes the cell to enter the S phase of the cell cycle, promoting DNA replication. Large T interacts with DNA polymerase-alpha to directly stimulate viral DNA replication. Anti-SV40T was created in 1981 to further understand the SV40 antigens via immunochemical analysis. It was thought that detailed mapping and analysis of the SV40 tumour antigens would provide information regarding the tertiary structure and ultimately the functions of these antigens. |
