Indirect enzyme linked immunosorbent assay (ELISA) using the anti-IL-22 monoclonal antibodies. A: ELISA performed using the anti-IL-22 antibody (clone L7) as the primary antibody. B: ELISA performed using the anti-IL-22 antibody (clone L8) as the primary antibody. 1: Untreated wells (negative control). 2: Wells coated with coating buffer only (negative control). 3: Wells coated with the IL-22 peptide immunogen #L7. 4: Wells coated with the IL-22 peptide immunogen #L8. 5: Wells coated with recombinant INFϒ. 6: Wells coated with recombinant IL-22.
Immunodetection of recombinant IL-22 by the monoclonal antibodies clones L7 and L8 respectively. MW: Molecular weight marker. Lane 1: Positive control (100 ng of recombinant IL-22). Lane 2: Negative control (100 ng of recombinant IL-2B). Both anti-IL22 antibodies (clones L7 and L8) are shown to specifically bind to recombinant IL-22.
Immunodetection of native IL-22 production within gill tissues after challenging with the bacterial fish pathogen Yersinia ruckeri. A: Relative expression of IL-22 in fish gill tissues as determined by qPCR. B: Immunodetection of IL-22 protein in the gill after purifying protein samples from Tri® Reagent. M: Molecular weight marker; +: Recombinant IL-22; -: Recombinant IL-2B. Fish number 1 exhibited low levels of IL-22 gene expression (A) and did not display detectable levels of IL-22 protein (B). Conversely, fish number 2 showed high levels of IL-22 gene expression (A) and produced detectable amounts of IL-22 protein (B). The MHC Class II antibody was used as a protein quality/loading control.
Immunohistochemistry was performed on formalin-fixed, paraffin-embedded gill tissue sections using Anti-IL22 [L8P2F10*A3]. Images are showing a strong immunostaining of IL22 in the gill tissues of Atlantic Salmon. Antigen retrieval step was required (microwave 10 min @ 950W in 0.01M sodium citrate buffer, pH 6.0).
Immunohistochemistry was performed on formalin-fixed, paraffin-embedded rainbow trout head kidney tissue sections. Clusters of IL22+ cells are showing strong positivity using Anti-IL22 [L8P2F10*A3]

Background and Research Application
IL-22 is an α-helical cytokine that binds to a heterodimeric cell surface receptor composed of IL-10R2 and IL-22R1 subunits. IL-22R is expressed on tissue cells, and it is absent on immune cells. IL-22 and IL-10 receptor chains play a role in cellular targeting and signal transduction to selectively initiate and regulate immune responses. IL-22 can contribute to immune disease through the stimulation of inflammatory responses, S100s and defensins.
