Relevance Conditional knockin of oncogenic B-Raf V600E; in vivo study of oncogenic B-raf mutation and signalling pathways The LSL-B-Raf-V600E targeting vector, containing a loxP flanked minigene+polyA encoding wildtype exons 15 to 18, amd a loxP flanked resistance cassette, and a substitution of residue 600 (V to E) in the endogenous exon 15, was transfected into E14.1 ES cells. Properly targeted ES cells containing homologous recombination events were injected into C57BL6 balstocysts. Chimeric mice were backcrossed on the C57BL6 background.
