Background and Research Application
CD45RO (UCHL1) is also known as protein tyrosine phosphatase receptor type C (PTPRC) or leukocyte common antigen. It is a transmembrane pan-leukocyte protein with tyrosine phosphatase activity and is involved in the regulation of signal transduction in haematopoiesis and the threshold of T cell antigen receptor (TCR) signalling. This is via dephosphorylation of protein tyrosine kinases (e.g. Lck and Fyn).
Anti-CD45RO subdivides the helper/inducer and suppressor/cytotoxic populations of T-cells, identifying sub populations of T cells which proliferate to soluble antigen and aid with PWM-stimulated immunoglobulin synthesis and B-cell differentiation.
CD45RO is the smallest isoform of the CD45 glycoprotein, produced via alternative splicing of exons 4-7, and is expressed on activated and memory T lymphocytes (around 50% of all T lymphocytes), most granulocytes and monocytes and most mature T cell tumours. CD45RO facilitates T cell activation, however, it is not present on B or NK cells.
CD45 regulates Src-family kinases, integrin-mediated signal transduction pathways and can suppress JAK kinases, acting as a negative regulator of cytokine receptor signalling.
CD45RO may respond via proliferation to recall antigens, provide help for secondary antibody responses and produce greater variety of cytokines.
This antibody reacts with an epitope unique for CD45R0. It may identify abnormalities of functional T-cell subsets in immune disorders, be used in tumour diagnosis by FACS or immunohistology and has been very useful in studies of human T cell function and ontogeny.

Points of Interest
Anti-CD45RO can differentiate between memory and naïve cells, which can be used to evaluate an immune response. It binds to immunising cultured T cells and may be used to identify abnormalities in functional T-cell subsets within immune disorders, offering a diagnostic function.
Previous uses of anti-CD45RO include tumour diagnosis via FACS or immune-histology, studies of human T cell function and ontogeny and identification of T vs B leukaemia.
Anti-CD45RO recognises a specific isoform present on a subset of cells, which can be used in diagnosis and identification of cell subsets.
Immunohistochemical staining has been undertaken with anti-CD45RO to evaluate T-lymphocytic immune reactions (e.g. in tumour microenvironment), in different disease states or cancer types.
