Monoclonal antibody directed against ErbB3 kinase, widely prevalent in GI and breast tumours. Background and Research Application The EGFR family of type I growth factor receptor tyrosine kinases includes EGFR (HER1), c-erbB2 (HER2; neu), c-erbB3 (HER3) and c-erbB4 (HER4). c-erbB3 is normally found in non-dividing differentiated epithelial cells, neurones and hepatocytes. ErbB3 was found to be kinase impaired, having only 1/1000 the autophosphorylation activity of EGFR and no ability to phosphorylate other proteins. Therefore, ErbB3 must act as an allosteric activator. The kinase-impaired ErbB3 is known to form active heterodimers with other members of the ErbB family, most notably the ligand binding-impaired ErbB2. It has approximately 80% prevalence in a wide variety of tumours of the gastro-intestinal tract, and approximately 20% prevalence in breast cancer. This antibody is specific against c-erbB-3 oncoprotein that is expressed in a variety of human normal, foetal and neoplastic epithelial tissues. |
Host | Mouse |
Immunogen | Synthetic Peptide from C terminus of human c-erbB3 |
Immunogen UniProt ID | P21860 |
Subclass | IgM |
Myeloma Used | NS0 |
Recommended Growing Conditions | RPMI 1640 + 10% FCS |
Notes |
Production Details Purified using multi-step affinity chromatography with protein A. Storage Conditions Store at -20 degrees frozen. Avoid repeated freeze/thaw cycles. Points of Interest This antibody can be used on formalin-fixed, paraffin-embedded tissue sections and frozen tissue sections. It reacts with normal gastrointestinal epithelium as well as gastrointestinal and breast tumours, resulting in a cytoplasmic staining pattern. Concentration 1mg/ml as standard |
