Info

Catalogue Number 151157
Applications IHC IP WB
Antigen/Gene or Protein Targets ErbB3 (Her3)
Reactivity Human
Relevance Monoclonal antibody directed against ErbB3 kinase, widely prevalent in GI and breast tumours.

Background and Research Application
The EGFR family of type I growth factor receptor tyrosine kinases includes EGFR (HER1), c-erbB2 (HER2; neu), c-erbB3 (HER3) and c-erbB4 (HER4). c-erbB3 is normally found in non-dividing differentiated epithelial cells, neurones and hepatocytes. ErbB3 was found to be kinase impaired, having only 1/1000 the autophosphorylation activity of EGFR and no ability to phosphorylate other proteins. Therefore, ErbB3 must act as an allosteric activator. The kinase-impaired ErbB3 is known to form active heterodimers with other members of the ErbB family, most notably the ligand binding-impaired ErbB2. It has approximately 80% prevalence in a wide variety of tumours of the gastro-intestinal tract, and approximately 20% prevalence in breast cancer. This antibody is specific against c-erbB-3 oncoprotein that is expressed in a variety of human normal, foetal and neoplastic epithelial tissues.
Host Mouse
Immunogen Synthetic Peptide from C terminus of human c-erbB3
Immunogen UniProt ID P21860
Subclass IgM
Myeloma Used NS0
Recommended Growing Conditions RPMI 1640 + 10% FCS
Notes Production Details
Purified using multi-step affinity chromatography with protein A.

Storage Conditions
Store at -20 degrees frozen. Avoid repeated freeze/thaw cycles.

Points of Interest
This antibody can be used on formalin-fixed, paraffin-embedded tissue sections and frozen tissue sections. It reacts with normal gastrointestinal epithelium as well as gastrointestinal and breast tumours, resulting in a cytoplasmic staining pattern.

Concentration
1mg/ml as standard
Research Area Cancer, Cell Type or Organelle Marker, Cell Signaling & Signal Transduction
