Monoclonal antibody which co-precipitates a2b1 integrin from keratinocytes and inhibits a2b1 motility in collagen. Background and Research Application Integrins are heterodimeric cell surface receptors composed of alpha and beta subunits, which mediate cell-cell and cell-extracellular matrix attachments. Aberrant integrin expression has been found in many epithelial tumours. Changes in integrin expression have been shown to be important for the growth and early metastatic capacity of melanoma cells. Integrin a2 associates with integrin ß1 (CD29) to form the heterodimer integrin a2ß1, which is one of the most abundant keratinocyte integrins, is found distributed in the basal layer of epidermis and binds collagen. Integrin alpha 2 beta 1 is the non-covalent heterodimer of 160 kDa alpha 2 (CD49b) and 130 kDa beta 1 (CD29) type I transmembrane glycoprotein subunits and is one of six very late antigens on activated T cells, designated VLA2. HAS-3 co-precipitates integrin a2ß1 from keratinocytes and inhibits HGF-induced branching morphogenesis and motility of human mammary epithelial HB2 cells in collagen. |

Production Details Purified using multi-step affinity chromatography with protein A. Storage Conditions Store at -20 degrees frozen. Avoid repeated freeze/thaw cycles. Points of Interest HAS-3 did not inhibit adhesion to laminin, fibronectin and type IV collagen. In innate immunity, alpha 2 beta 1 binding to C1q initiates the complement cascade and co-stimulates mast cell activation, triggering neutrophil influx. Concentration 1mg/ml as standard |
