EPAS-1 Antibody (190b) is an IgG1 κ mouse monoclonal EPAS-1 antibody (also designated EPAS1 antibody, HIF2A antibody, or HIF-2-alpha antibody) that detects the EPAS-1 protein of mouse, rat and human origin by WB, IP, IF, IHC(P) and ELISA. EPAS-1 Antibody (190b) is available as both the non-conjugated anti-EPAS-1 antibody form, as well as multiple conjugated forms of anti-EPAS-1 antibody, including agarose, HRP, PE, FITC and multiple Alexa Fluor® conjugates. Cell growth and viability is compromised by oxygen deprivation (hypoxia). Hypoxia-inducible factors, including HIF-1α, HIF-1β (also designated Arnt 1), EPAS-1 (also designated HIF-2α) and HIF-3α, induce glycolysis, erythropoiesis and angiogenesis in order to restore oxygen homeostasis. Hypoxia-inducible factors are members of the Per-Arnt-Sim (PAS) domain transcription factor family. In response to hypoxia, HIF-1α is upregulated and forms a heterodimer with Arnt 1 to form the HIF-1 complex. The HIF-1 complex recognizes and binds to the hypoxia responsive element (HRE) of hypoxia-inducible genes, thereby activating transcription. Hypoxia-inducible expression of some genes such as Glut-1, p53, p21 or Bcl-2, is HIF-1α dependent, whereas expression of others, such as p27, GADD 153 or HO-1, is HIF-1α independent. EPAS-1 and HIF-3α have also been shown to form heterodimeric complexes with Arnt 1 in response to hypoxia.

What is recommended incubation time for western blot of cell extracts with sc-13596? Thank You.

Thank you for your question. We recommend incubating the primary antibody for 1 hour at room temperature (or overnight at 4C) in 5% nonfat milk in TBST.

When should the TransCruz reagent sc-13596 X be used instead of the standard form sc-13596? What is the difference between the two forms?

Thank you for your question. We recommend using the TransCruz reagent form when performing ChIP or GelShift assays. This form can also be used in other applications such as Western Blot or IHC, but the dilution should be adjusted by a factor of 10 to account for the higher original concentration. The TransCruz reagent is provided as 10x more concentrated than the regular form: sc-13596 X, 200 µg/0.1 ml.

Non ho rilevato alcuna banda nel caso delle C28I2, mentre nelle H9c2 compare una banda ma ad un peso molecolare molto diverso da quello atteso (circa 45-50 kDa).
