CD14 Antibody (UCH-M1) is an IgG2a κ mouse monoclonal CD14 antibody that detects CD14 monocyte specific cell surface protein of human origin by WB, IP, IF, IHC(P) and FCM. CD14 Antibody (UCH-M1) is available as the non-conjugated anti-CD14 antibody form, as well as multiple conjugated forms of anti-CD14 antibody, including agarose, HRP, PE, FITC and multiple Alexa Fluor® conjugates. Lipopolysaccharide (LPS) elicits the secretion of mediators and cytokines produced by activated macrophages and monocytes. CD14 is a glycosylphosphatidylinositol (GPI)-anchored protein found on the surfaces of monocytes and polymorphonuclear leukocytes. CD14 functions as a receptor for LPS, resulting in the secretion of various proteins. An important component in the LPS activation of monocytes through the CD14 receptor is the "adapter molecule," lipopolysaccharide binding protein (LBP). There are two forms of CD14, a membrane-associated form (mCD14), and a soluble form (sCD14). mCD14 responds to LPS alone and facilitates the secretion of proteins, while cells not expressing mCD14 fail to respond to LPS. The cells that lack mCD14 respond to LPS/LBP in the presence of sCD14.

What is the CD14 concentration used for flow cytometry?

Asked by: Bravo_biologist
Thank you for your question. You can find our recommended protocol for flow cytometry under this link: https://www.scbt.com/resources/protocols/flow-cytometry. We recommend using 1 µg of fluorochrome-conjugated antibodies per 1 million cells in 100 µl buffer per tube.

What negative control should be used when using the conjugated version of CD14 (UCH-M1): sc-1182 monoclonal antibody in flow cytometry?

Asked by: jenniferc
Thank you for your inquiry. We recommend using the isotype specific normal mouse IgG conjugated to the same fluorophore. For example, if you are using sc-1182 PE, then you would want to use normal mouse IgG2a PE, sc-2867.

What secondary is best to use with CD14 (UCH-M1): sc-1182 monoclonal antibody?

Asked by: jenniferc
Thank you for your question. While most anti-mouse IgG secondary antibodies are appropriate for use with CD14 (UCH-M1): sc-1182, we recommend using one our exclusive Mouse IgG Binding Proteins as a secondary detection reagent. A complete list of available binding proteins is available on our website here: https://www.scbt.com/scbt/browse/support-products-mouse-igg-binding-proteins/_/N-ecrety
