PCNA Antibody (PC10) is an IgG2a κ mouse monoclonal PCNA antibody (also designated ATLD2 antibody) that detects the PCNA protein of m, r, h, insect, S. pombe origin by WB, IP, IF, IHC(P) and FCM. PCNA Antibody (PC10) is available as both the non-conjugated anti-PCNA antibody form, as well as multiple conjugated forms of anti-PCNA antibody, including agarose, HRP, PE, FITC and multiple Alexa Fluor® conjugates. The PCNA (proliferating cell nuclear antigen) protein is synthesized in early G1 and S phases of the cell cycle, functions in cell cycle progression, DNA replication and DNA repair. In early S phase, the PCNA protein exhibits granular distribution and is absent from the nucleoli; however, in late S phase, it relocates to the nucleoli. Thus, in staining applications, the PCNA Antibody exhibits nuclear staining. PCNA exists in two basic forms: one involved in ongoing DNA replication, which localizes specifically to the nucleus, and a second, soluble form, not implicated in constant synthesis. Interestingly, the latter form degrades in the presence of organic solvents, rendering it undetectable by histological methods in tissues using organic fixatives. Thus, only the synthesizing form is detectable by the PCNA antibody using this method.

does antibody m-IgGκ BP-HRP:(sc-516102) compatible to using as secondary antibody against Anti-PCNA Antibody (PC10): sc-56 for western blot

Thank you for your question. Anti-PCNA Antibody (PC10) is a mouse monoclonal IgG2a κ PCNA antibody, so it is compatible with m-IgGκ BP-HRP:(sc-516102), which recognizes kappa light chain mouse IgG, IgA and IgM antibodies.

Recently we observed that the PCNA sc-56 antibody in the tube is precipitated. It was stored at 4 degrees as recommended. Can the antibody be used for experimental purpose? If yes how do we use it? any recommendation from company's side?

Which antigen retrieval buffer should be used with PCNA sc-56, citrate or Tris? Can you please include this information in the product datasheet so that it becomes convenient for the user to begin with.

Thank you for your question. We recommend using sodium citrate buffer (pH 6.0) with heat treatment for antigen retrieval with this PCNA antibody. The detailed method can be found in our recommended protocol online.

Can I use this mouse monoclonal antibody for immunohistochemical staining of swine(pig) tissue?

Thank you for your question. We have not tested this antibody on swine or pig tissue, however, we are aware of a few publications citing use of this antibody in pig samples. For example, PMID: 26389662.

We are new to IHC. Thinking of using this antibody, PCNA Antibody: sc-56on our chicken samples. Wondering if there are any preferences on the conjugate for the secondary antibody, such as HRP, biotin etc. We don't need fluorescence

Thank you for your question. This antibody has been previously cited for detection of PCNA in chicken samples. For IHC, you could use the unconjugated antibody, with an appropriate secondary antibody for detection, or the Biotin conjugated primary with an ABC Staining System, or the HRP conjugate for direct immunoperoxidase staining. Using the directly conjugated primary antibody can save you time and money on reagents. We do not have a preference for this particular antibody, and think you would be successful using either version of the antibody, depending on your preferences in your lab.

I'm wondering about the region of PCNA that is recognized by this antibody SC-56, PC10? Would an N-terminal Flag tag get in the way? Thank you.

Thank you for your question. The exact epitope recognized by this mouse monoclonal antibody has not been determined. However, some report that this clone (PC10) recognizes an epitope within the amino acid range of 111-125. In that case, we would not expect an N-terminal Flag tag to prevent this antibody from recognizing PCNA that contains this sequence.
