ICAM-1/CD54 Antibody (15.2) is an IgG1 κ mouse monoclonal ICAM-1 antibody that detects ICAM-1 of mouse, rat and human origin by WB, IP, IF, IHC(P) and FCM. ICAM-1/CD54 Antibody (15.2) is available as the non-conjugated anti-ICAM-1 antibody form, as well as multiple conjugated forms of anti-ICAM-1 antibody, including agarose, HRP, PE, FITC and multiple Alexa Fluor® conjugates. Cell adhesion molecules (CAMs) are a family of closely related cell surface glycoproteins involved in cell-cell interactions during growth and are thought to play important, yet separate, roles in embryogenesis and development. The intracellular adhesion molecule-1 (ICAM-1), also referred to as CD54, is an integral membrane protein of the immunoglobulin superfamily and recognizes the β2α1 and β2αM Integrins. ICAM-2 functions as a ligand for lymphocyte function-associated antigen-1 (LFA-1) and is involved in leukocyte adhesion. ICAM-3 is highly expressed on the surface of human eosinophils and, when bound to ligand, may inhibit eosinophil inflammatory responses and survival. ICAM-4, also known as LW glycoprotein, interacts with Integrins αLβ2, αMβ2, α4β1, the αV family and αIIbβ3, and selective binding to different integrins may be relevant to the pathology in a number of red blood cell associated diseases. Lastly, ICAM-5, expressed on telencephalic neurons, binds CD11a/CD18 and thus may act as an adhesion molecule for leukocyte binding in the central nervous system.

Thank you for your question. We recommend using sodium citrate buffer (pH 6.0) with heat treatment for the antigen retrieval method when using this antibody on formalin-fixed paraffin-embedded tissue sections, or for IHC(P). How many Flowcytometry and IHC tests could be conducted with your supplied antibody reagents for ICAM1, VE cad and ZO1? Its urgent so please let me know asap!

Thank you for your question. The recommended starting dilution for IHC is 1:50 (range of 1:25-1:500). The number of slides that can be stained depend on your dilution, as well as your working volume.

What is the expected band size for ICAM-1 using sc-107 for western blot?

Thank you for your question. The expected molecular weight for ICAM-1 is 85-110 kDa due to glycosylation. There is representative Western blot data on our website for this specific antibody.

What negative control should be used when using the conjugated version of this antibody in flow cytometry?

Thank you for your inquiry. We recommend using the isotype specific normal mouse IgG conjugated to the same fluorophore. For example, if you are using sc-107 PE, then you would want to use normal mouse IgG1 PE, sc-2866.
