Genetic informationThe genomic DNA encoding the ATP-binding DFG motif in the Pik3c2a (PI3K-C2alpha) gene is mutated to encode the AFG sequence (amino acid substitution of alanine for aspartic acid at position 1268, D1268A), resulting in the production of a kinase-dead PIK3C2A protein.
Phenotypic informationHomozygous:
Homozygous mice are lethal between embryonic day 10.5 and 11.5

Heterozygous:
Heterozygous Pik3c2a mutant mice were viable and fertile, with no significant histopathological findings. However, male heterozygous mice showed early onset leptin resistance, with a defect in leptin signalling in the hypothalamus, correlating with a mild, age-dependent obesity, insulin resistance and glucose intolerance. Insulin signalling was unaffected in insulin target tissues of Pik3c2a mutant mice. No metabolic phenotypes were detected in female Pik3c2a mutant mice at any age.
Breeding historyBackcrossed onto C57BL/6J for more than 10 generations.
