  • Additional notes: The pETHSUL vector is part of a set of ligation independent cloning SUMO fusion vectors that allow for recombinant proteins to be produced with any desired sequence at the amino terminus, so long as the initial residue is not proline. This set of vectors generates affinity tagged SUMO fusion target proteins that allow the target protein to be expressed in E.coli and purified using subtractive affinity chromatography. The purified SUMO fusion is treated with recombinant SUMO protease (generated by pdtUD1 vector) to yield the mature target protein. The pETHSUL vector yields His6 tagged SUMO fusion proteins. Recombinant SUMO protease cleaves His6 SUMO from the purified target protein to yield the final mature protein. The pETHSUL vector was constructed based on the pET vector architecture.
