Cell Line Description

Adherent keratinocyte cell line derived from a squamous cell carcinoma of the alveolus of a Caucasian female. Keratin and involucrin markers present. Known mutations: p53 and p16. Cells are tumourigenic in athymic mice. Epidermal Growth Factor Receptor (EGFR) present.

Subculture Routine

Split sub-confluent cultures (70-80%) 1:2 using 0.05% trypsin/EDTA; 8% CO₂; 37°C. Suggested seeding density 4 x 10,000 cells/cm². A feeder layer of lethally-irradiated or mitomycin C-treated 3T3 feeder cells can be used to culture this cell line, however, this is not essential. The cell line was cultured at ECACC in the absence of a feeder layer. Use of conditioned medium at a ratio of 1:1 with fresh medium can improve cell condition.

Culture Medium

DMEM + 2mM Glutamine + 10% Foetal Bovine Serum (FBS) + 0.4 µg/ml Hydrocortisone.
