Cell Line Description

The stable human mammary MCF7-derived reporter cell line called AREc32 contains copies of the rat Glutathione-S-transferase (GST) antioxidant response element (ARE) linked to a luciferase gene, such that the induction of the ARE results in luciferase activity. ARE is a transcriptional regulatory element involved in the activation of genes coding for a number of antioxidant proteins and enzymes that work in concert to protect tissues from oxidative insults. This cell line can be used to investigate whether anticancer drugs can induce ARE-driven gene expression.

Split sub-confluent cultures (70-80%) 1:3 to 1:6 i.e. seeding at 2-4x10⁴ cells/cm²using 0.05% trypsin/EDTA solution. Cells are cultured at 37°C, in 95% air and 5% CO₂, and passaged every 3 to 4 days

Dulbecco's Moodifies Eagle Medium (DMEM) with 2.5 mM Glutamax supplemented with 10% foetal bovine serum. Do not culture beyond 15 passages after revival
