Background and Research Application
CD45RB is an isoform of CD45 with exon 5 splicing (encodes B determinant). It is a 220 kD glycoprotein expressed on peripheral B cells, naïve T cells, thymocytes, weakly on macrophages, and dendritic cells. CD45 regulates Src-family kinases, integrin-mediated signal transduction pathways and can suppress JAK kinases, acting as a negative regulator of cytokine receptor signalling. It plays a critical role in TCR and BCR signalling. As T cells become activated and progress from naïve to memory cells, CD45RB expression is downregulated. CD45RB is expressed in microglia and inflammatory cells. In addition to involvement in immunoreceptor signalling, CD45 is important in promoting cell survival by modulating integrin-mediated signal transduction pathway and is involved in DNA fragmentation during apoptosis.
