Background and Research Application
The murine 5T4 monoclonal antibody, clone B3F1, identifies the human 5T4 oncofoetal antigen, also known as trophoblast glycoprotein. 5T4 is a highly glycosylated protein expressed by trophoblast and a few specialized adult epithelia and is an antagonist of the Wnt/ β-catenin signalling pathway.
Up-regulation of 5T4 expression in some cancers is associated with poor clinical outcome; overexpression of human 5T4 cDNA in epithelial cells can alter their morphology and motility, supporting a role for such functions in cancer and development. 5T4 has limited expression in normal tissues, but is widespread in tumours, thus its expression is used as a prognostic aid. 5T4 is typically found in colorectal, ovarian and gastric tumours.
5T4 is a potential target for T cells, in antibody-directed immunotherapy.

Recommended Growing Conditions DMEM supplemented with heat inactivated 10% fetal calf serum, 2 mM L-glutamine, 100 units/ml penicillin and 100 µg/ml streptomycin solution and maintained at 37°C in a humidified atmosphere of 5% CO2/95% air.

Points of Interest
A study found that 5T4 was present in 85% of a study of 72 colorectal carcinomas and in 81% of a study of 27 gastric carcinomas.

Clone B3F1 inhibits the chemotactic migration towards CXCL12 exhibited by differentiating WT-ES cells (to a lesser extent than clone B1C3). IC50 for the maximally inhibitory mAb B3F1 was 2.2 µg±0.8.
