Relevance Herpes simplex virus (HSV) virions are multilayered, and their assembly requires several steps (reviewed in references 21 and 59). The double-stranded DNA viral genome is enclosed within a well-ordered protein capsid. A more amorphous layer of proteins referred to as the tegument surrounds the capsid. Lying outside of the tegument, the outermost layer of the virion consists of a lipid envelope containing viral glycoproteins. Herpes simplex virus type 1 (HSV-1) capsids are initially assembled with an internal protein scaffold. The scaffold proteins, encoded by overlapping in-frame UL26 and UL26.5 transcripts, are essential for formation and efficient maturation of capsids.
