Relevance Monoclonal antibody which binds anti-apoptotic regulator BCL2, used to aid in differentiating between various lymphomas and as prognostic biomarker in various cancers.

Background and Research Application
BCL2 is a member of the BCL-2 family of regulatory proteins, involved in apoptosis, which promotes cell survival and inhibits the role of pro-apoptotic proteins such as Bax and Bak. The pro-survival protein BCL2 derives its name from B-cell lymphoma 2. It is the second member of a range of proteins initially described in chromosomal translocations involving chromosomes 14 and 18 in follicular lymphomas. Damage to the Bcl-2 gene has been identified as a possible cause of schizophrenia and autoimmunity, a cause of cancers, including melanoma, breast, prostate, chronic lymphocytic leukaemia, and lung cancer. It also provides resistance to cancer treatments .
This antibody was created to see if the BCL2 protein is a specific marker to the 14;18 chromosomal translocation in human follicular lymphoma. Previous studies claimed the BCL2 protein was not found in normal tissue but was present in all lymphomas where this specific translocation was observed. This antibody helped show the immunohistologic demonstration of BCL2 isn’t specific for the presence of 14;18 translocation.

Points of Interest
Anti-BCL2 can be used to differentiate between lymphomas which express BCL2 and the small proportion of neoplasms which are BCL2 negative, and has been used as a predictive biomarker for recurrence of radical prostatectomy for prostate cancer.
Anti-BCL2 labels neoplastic lymphoid follicles. In the case of follicular lymphoma, its value is to distinguish between reactive and neoplastic follicular proliferation in lymph node biopsies. It can also help differentiate between follicular lymphoma and diffuse large cell lymphomas, and between follicular lymphomas and reactive lymph nodes.
