Relevance Monoclonal antibody which recognises circulating blood monocytes and the large majority of tissue macrophages, without labelling granulocytes or lymphocytes.

Background and Research Application
Anti-haemoglobin scavenger receptor labels the majority of monocytes and tissue macrophages. It recognises a membrane glycoprotein with molecular weight 130,000 (reduced) and 110,000 (unreduced). The glycoprotein antigen is expressed exclusively on the cell surface of human monocytes and macrophages, which evolves predominantly in the late phase of inflammation. The glycoprotein GHI/61 binds to CD163, a transmembrane glycoprotein belonging to the scavenger receptor cysteine-rich superfamily. It is a receptor for the haemoglobin-haptoglobin complex.
CD163 expression on monocytes and macrophages changes as these cells differentiate, as such GHI/61 can detect levels of differentiated monocytes and macrophages. CD163 has high affinity for the haemoglobin-haptoglobin complex and upon ligand binding, leads to the production of IL-6 and IL-10.

GHI/61 monoclonal antibody was created to label monocytes and macrophage with increased specificity, given similar antibodies frequently label lymphocytes etc., or only recognise a particular subset of macrophages.

Points of Interest
GHI/61 does not label lymphocytes, which is a drawback of other monocyte labelling antibodies. GHI/61 only reacted with normal monocytes and macrophages when tested upon a variety of leukaemia’s and lymphomas. All monocytes were positive for FACS staining following permeabilization, with no lymphocytes or granulocytes stained, giving more accurate results. The absence of granulocyte (and its precursors) staining may be of value within leukaemia and lymphoma diagnosis.

GHI/61 antibody recognises the same antigen as Ber-MAC3, Ki-M8 and SM4. However, it seems all four label different epitopes of the same antigen.
Labelling of the tonsil indicated GHI/61 didn’t recognise interdigitating reticulum cells, dendritic reticulum cells or Langerhans cells.

Ultroser G can be used at 1% if the cells are not growing well.
